The surface point you want to dive in from is the Drenan Highlands Chasm in the Thyphlo Ruins part of the map. That’s the very top and center area of the map past Hyrule castle, and past the ominous purple island.

Once you’ve dropped down through that chasm, you need to go northeast. Mostly east. The Den’s entrance is just a short walk from the Chasm.

If you’ve rather not navigate in pure darkness, the nearest Lightroot is the Nikakik Lightroot. To get to it, turn around and go southwest for a while. If you want to be able to actually see the Gleeok Den on the map first, then you have to go north and then east from the chasm. This will let you reach the Katenim Lightroot above the den. Having this Lightroot will make the Gleeok Den visible so you’ll always know where to find the Gleeok Den in Zelda: Tears of the Kingdom for the future.

Well, at least if you don’t, it’s not a long trek back if you go up to the surface for supplies and come back.
